Prior to 2009, we built our reputation on providing high-quality reagents for molecular biology applications. The next-generation sequencing revolution was gaining momentum, and we took notice!
In 2009, we debuted the NEBNext® portfolio of sample prep products for NGS, with the goal of making NGS more accessible to a broad range of scientists.
Our first NGS products included:
• Reagent kits and modules for DNA library prep
• A new solution for enzyme-based DNA fragmentation
• Options for users of Illumina® or 454® sequencing platforms
We next added RNA library prep solutions for Illumina sequencing to the NEBNext product line and began supporting library prep for the SOLiD™ sequencing platform.
As a time-saving step towards streamlined workflows, NEBNext Master Mix Kits were introduced, which minimized components, protocol steps and complexity.
Adding to the list of supported platforms, NEBNext products for Ion Torrent® sequencers and kits to enable ChIP-seq library prep for Illumina sequencing were added to the portfolio.
A novel solution to the challenge of adaptor-dimer formation in Small RNA library prep inverted the typical adaptor hybridization and ligation steps, substantially minimizing adaptor-dimers.
We then introduced the Ultra™ product line for DNA and RNA Illumina sequencing, combining enzymatic reactions to streamline and simplify workflows. Input amounts were reduced 200-fold, clean-up steps were minimized, and library quality and yields grew. These advances made Ultra library prep the method of choice, even for those using precious samples in scarce supply.
Other advances included introduction of the hairpin-loop structure NEBNext Adapter, which improved ligation efficiency, and release of our first PCR Master Mix optimized for amplification of NGS libraries.
As the microbiome’s important role in health and illness was first coming to light, researchers required new, reliable methods to work with these sample types. To that end, we released the NEBNext Microbiome DNA Enrichment Kit to separate microbial DNA from host genomic DNA.
A major impediment to RNA sequencing is the prevalence of ribosomal RNA (rRNA) species that can monopolize an RNA-seq run. To address this, we introduced the RNase H-based NEBNext rRNA Depletion Kit, for more cost-effective sequencing of both high and low quality RNA.
To support the increase in sample multiplexing, we also released our first set of 96 dual-index primer pairs for improved sample identification specificity with Illumina library production.
NEBNext Ultra II DNA Library Prep reagents reduced inputs an additional 10-fold, while improving efficiency at each step and enabling use with even more challenging samples.
To support sequencing of fixed and archived tissue samples, the NEBNext FFPE DNA Repair Mix was introduced to repair several of the most common types of DNA damage present in FFPE samples.
For accurate quantitation and standardization of libraries prior to a sequencing run, our Library Quant Kit uses a qPCR-based workflow and accurate standards to count only molecules that contain both adaptor sequences.
Targeted genome analysis by sequencing only regions of interest is supported by our NEBNext Direct® target enrichment product line. The first product released was the Cancer Hotspot Panel - a preselected panel of 190 targets across 50 cancer-related genes.
2016 was also the year we introduced 96-well plate format index primers for multiplexing.
DNA fragmentation had been a challenging part of the library prep workflow. As a new enzymatic alternative to mechanical shearing methods, and with a requirement for reliability and simple scalability, the NEBNext Ultra II FS DNA library prep products were launched. The FS enzyme mix fragments, end repairs and dA-tails input DNA without cut-site bias and with a simple protocol effective with as little as 100 pg DNA.
Continuing to meet demand for streamlined workflows and lower inputs, Ultra II RNA library prep kits reduced input amount minimums 10-fold, at the same time increasing library quality and yields.
Releasing several new products each year, the NEBNext product line continued to grow. 2018 saw our entry into the single cell space with the NEBNext Single Cell/Low Input RNA products, incorporating a unique workflow for full-length transcript detection.
We also addressed speed for customized target enrichment with a new
product format: NEBNext Direct Custom Ready, with the ability
to create customized panels of pre-designed targets with a
two week turnaround time.
Now, more than 60 NEBNext products are available to propel your next-generation sequencing research.
Recent releases include:
• NEBNext Enzymatic Methyl-seq (EM-Seq™) — a gentler alternative to sodium bisulfite sequencing, for a more effective methylome analysis.
• The NEBNext Globin & rRNA Depletion Kit solves the problem of RNA sequencing from blood samples by selective removal of the predominant RNA species present, globin mRNA and rRNA.
• The NEBNext Companion Module for Oxford Nanopore Technologies® Ligation Sequencing — providing the NEBNext reagents for use alongside SQK-LSK109 in just the amounts needed.
